== C-terminal amino acid analysis == The number of methods available for C-terminal amino acid analysis is much smaller than the number of available methods of N-terminal analysis. The most common method is to add carboxypeptidases to a solution of the protein, take samples at regular intervals, and determine the terminal amino acid by analysing a plot of amino acid concentrations against time. This method will be very useful in the case of polypeptides and protein-blocked N termini. C-terminal sequencing would greatly help in verifying the primary structures of proteins predicted from DNA sequences and to detect any posttranslational processing of gene products from known codon sequences.

== Martin and Synge and partition chromatography == Chromatography methods changed little after Tsvet's work until the explosion of mid-20th-century research in new techniques, particularly thanks to the work of Archer John Porter Martin and Richard Laurence Millington Synge. By "the marrying of two techniques, that of chromatography and that of countercurrent solvent extraction", Martin and Synge developed partition chromatography to separate chemicals with only slight differences in partition coefficients between two liquid solvents. Martin, who had previously been working in vitamin chemistry (including attempts to purify vitamin E), began collaborating with Synge in 1938, brought his experience with equipment design to Synge's project of separating amino acids. After unsuccessful experiments with complex countercurrent extraction machines and liquid-liquid chromatography methods where the liquids move in opposite directions, Martin hit on the idea of using silica gel in columns to hold water stationary while an organic solvent flows through the column. Martin and Synge demonstrated the potential of the methods by separating amino acids marked in the column by the addition of methyl red. In a series of publications beginning in 1941, they described increasingly powerful methods of separating amino acids and other organic chemicals. == Side effects == The side effects of EMP overall have been described as relatively severe. The most common side effects of EMP have been reported to be gastrointestinal side effects like nausea, vomiting, and diarrhea, with nausea and vomiting occurring in 40% of men. They are usually mild or moderate in severity, and the nausea and vomiting can be managed with prophylactic antiemetic medications. Nonetheless, severe cases of gastrointestinal side effects with EMP may require dose reduction or discontinuation of therapy. Although nausea and vomiting have been reported to be the most common side effects of EMP, gynecomastia (male breast development) has been found to occur in as many as 83% of men treated with EMP, and the incidence of erectile dysfunction is possibly similar to or slightly less than the risk of gynecomastia. As a rule, feminization, a gynoid fat distribution, demasculinization, and impotence are said to occur in virtually or nearly 100% of men treated with high-dose estrogen therapy. Decreased sexual activity has also been reported in men treated with EMP. These side effects are due to high estrogen levels and low testosterone levels. Prophylactic irradiation of the breasts can be used to decrease the incidence and severity of gynecomastia with estrogens.
